My research focuses on the investigation of proteolytic enzymes, primarily deubiquitinases such as the papain-like protease (PLpro) of SARS-CoV-2. I develop and validate chemical tools (e.g. fluorogenic substrates and activity-based probes) which are highly selective towards SARS-CoV-2 PLpro. Such tools make it possible to study the enzyme in detailed way and thereby to get a deeper understanding of its role during infection. By utilizing activity-based probes, I seek to develop an assay that will accelerate the screening of potential SARS-CoV-2 PLpro inhibitors.
This work combines many areas of chemistry and biology and thus I employ various techniques originating from these fields, including solid-phase peptide synthesis, enzyme kinetic studies, LC-MS analysis, cell-based assays, SDS-PAGE, western blotting and more.
